摘要 | The two primary modalities of vibrational spectroscopy, IR and Raman spectroscopy, provide complementary and comprehensive chemical and molecular information. Whereas Fourier-transform infrared (FTIR) microscopy suffers from low spatial resolution and strong water absorption background, the emerging technique of optical photothermal infrared (O-PTIR) microscopy is introduced with the advantages of reaching the spatial resolution at the nanometer scale. More recently, O-PTIR microscopy has been integrated with Raman spectroscopy, which offers submicron infrared photothermal images and simultaneous infrared and Raman spectra that complement each other. This chapter discusses the working principle, instrumentation, and performances of the Raman-integrated O-PTIR microscopy. Raman-integrated O-PTIR microscopy has high application potential, especially in the field of biomedicine. The latest applications include characterization of lipid droplets in single cells, fingerprinting a single bacterium, and phenotyping bacterial responses to drugs. Applications in the chemical and material analysis are also presented. |
