摘要	目的研究Toll样受体4(TLR4)和脂多糖受体(CD_(14))基因多态性在温州地区汉族儿童中的分布特征及其与特应性疾病的关系.方法 2004-02-2006-04,于温州育英儿童医院,利用测序法测定79例健康对照组儿童和113例特应征儿童的TLR4基因Asp299Gly和Thr399Ile的基因序列以及CD_(14)基因序列,寻找多态位点,调查多态位点的分布特征,比较两组儿童多态位点的基因型频率和等位基因频率.结果 79例对照组儿童和113例特应征组儿童均未发现TLR4基因Asp299Gly和Thr399Ile多态性.特应征组和对照组儿童均发现CD_(14)/-159多态性,以TT基因型为主,未发现其他多态位点.其中对照组儿童TT、TC、CC 3种基因型频率分别为57.0%、28.0%和15.0%,特应征组儿童TT、TC、CC 3种基因型频率分别为46.9%、35.4%和17.7%,按Hard-Weiberg平衡吻合度检验,差异无显著性(x~2=3.462,P＞0.05).两组的基因型频率和等位基因频率分布差异无显著性(x~2=1.918,P＞0.05).结论未发现TLR4基因Asp299Gly和Thr399Ile多态性,温州地区汉族儿童存在CD_(14)/-159多态性,未发现CD14基因其他多态位点.温州地区汉族儿童CD_(14)/-159基因型以TT为主,未发现CD_(14)/-159基因型和特应征发病的相关性.
其他摘要	Objective To investigate the distribution characteristics of the single nucleotide polymorphisms ( SNPs) of the human Toll-like receptor 4 ( TLR4 ) gene and CD_(14) gem; in Chinese Han children in Wenzhou, and their relation to atopic diseases. Methods A total of 113 cases diagnosed as atopic disease and 69 healthy children were enrolled. The related region of TLR4 gene containing Asp299Gly and Thr399Ile polymorphisms and CD_(14) gene were sequenced to identify and characterize the SNPs. The frequency of genotypes and ulleles between two groups were compared. Results We did not find Asp299Gly and Thr399Ile polymorphisms in the loll-like receptor 4 gene in both groups; CD_(14)/- 159 SNP was present in Han population of Wensthou. The frequency of each genotype was 57.0% (TT), 28.0% (TC) ,15. 0% ( CC) in normal children, and 46. 9% (TT),35.4% (TC), 17.7% (CC) in atopic children. No significant difference was found in the distribution of CD_(14)/- 159 polymorphism between atopic children and healthy controls (x~2 =1.918, P >0. 05) according to Hardy-Weinberg principle statistics. Conclusion Asp299Gly and Thr399Ile polymorphisms in the toll-like receptor 4 gene are not common, in Han children in Wenzhou. CD_(14)/ -159 SNP is present in Han children in Wenzhou, and other SNP in CD_(14) gene is not found. TT genotype is the primary genotype in CDl4/ - 159 SNP in Han children in Wenzhou. No relationship between CD_(14)/- 159 SNP and atopic disease is found.
