摘要 | 目的: 研究银杏叶提取物(EGB)对I 型糖尿病心肌病大鼠心肌TGF-β_1和collagen 表达及间质纤维化的影响。方法: (1)在体实验,取30只雄性SD 大鼠随机分为正常对照组(CON 组) 、糖尿病组(DM 组)和EGB干预组(EGB 组) ; 用链脲佐菌素诱导I 型糖尿病大鼠模型,于第12周末测定大鼠体重、左心室重量、血糖、糖化血红蛋白和血胰岛素; 用超声心动图仪检测左室舒张末期容积(LVEDV) 、左室收缩末期容积(LVESV) 、左室射血分数(LVEF)和每搏射血量(SV) ; 用天狼星红染色、免疫组织化学染色及RT-PCR 方法分别检测左心室心肌胶原含量、转化生长因子β_1(TGF-β_1)蛋白表达及TGF-β_1、procollagen I 和collagen III mRNA 表达。(2)离体实验,分离培养SD 大鼠乳鼠心肌细胞并分别用低糖(LG 组) 、高糖培养基(HG 组)和EGB 干预(HG + EGB 组) ; RT-PCR 方法检测各组心肌细胞TGF-β_1、procollagen I 和collagen III 的mRNA 表达。结果: (1)与CON 组相比,DM 组大鼠血糖(P < 0. 01) 、糖化血红蛋白(P < 0. 05) 、左心室重量指数(P < 0. 01) 、心肌间质胶原纤维含量(P < 0. 05)以及心肌组织内TGF-β_1(P < 0. 05) 、procollagen I(P < 0. 05)和collagen III(P < 0. 05)达均明显升高,而血胰岛素水平(P <0. 01) 、LVEDV (P < 0. 01) SV (P < 0. 01)明显降低; EGB 干预治疗后,与DM 组相比,EGB 组糖尿病大鼠血糖(P < 0. 01) 、糖化血红蛋白(P < 0. 05) 、左心室重量指数(P < 0. 05) 、心肌间质胶原纤维含量(P < 0. 05)心肌组织内TGF-β_1(P < 0. 05) 、procollagen I(P < 0. 05)和collagen III(P < 0. 05)表达均明显降低,血胰岛素水平(P <0. 05) 、LVEDV (P < 0. 05)和SV (P < 0. 05)明显升高。(2)与LG 组比较,HG 组心肌细胞TGF-β_1(P < 0. 01) 、procollagen I(P < 0. 01)和collagen III(P < 0. 01) mRNA 表达明显升高; 而EGB 干预后,与HG 组比较,EGB 组心肌细胞TGF-β_1(P < 0. 01) 、procollagen I(P < 0. 01)和collagen III(P < 0. 01) mRNA 表达明显降低。结论: EGB 可通过调低糖尿病大鼠心肌组织TGF-β_1的表达,减少心肌间质中collagen I 和III 合成及沉积,从而改善糖尿病心肌病大鼠早期的心肌纤维化及左心室舒张功能。这提示通过有效降低心肌TGF-β_1的表达减少心肌间质纤维化可能是EGB防治糖尿病心肌病的重要机制之一。 |
其他摘要 | AIM: To study the effects of Ginkgo biloba extract (EGB) on myocardial TGF-β1 and collagen expression and interstitial fibrosis in type I diabetic cardiomyopathy rats. METHODS: Thirty male SD rats were randomly divided into normal control group (CON),diabetes mellitus group (DM) and EGB treatment group (EGB). Streptozocin was intraperitoneally injected into the animals in the latter 2 groups to induce type I diabetic rat model. The rats in EGB group were intraperitoneally injected with EGB. At the end of the 12th week,the body weight of each rat and its left ventricular weight,blood glucose,glycosylated hemoglobin and serum insulin concentration were measured. The left ventricular end-diastolic volume (LVEDV),the left ventricular end-systolic volume (LVESV),the left ventricular ejection fraction(LVEF) and the stroke volume (SV) were determined by echocardiography. The content of collagen in left ventricular myocardium,and the expression of transforming growth factor β_1(TGF-β_1),procollagen type I and collagen type III were assayed by Sirius red staining,immunohistochemical staining and RT-PCR,respectively. Left ventricular myocardial cells of the neonatal SD rats were isolated and cultured in vitro with low-glucose culture medium (LG group),high-glucose culture medium (HG group) or high-glucose culture medium plus EGB (HG + EGB group). The mRNA levels of TGF-β_1, procollagen type I and collagen type III were detected by RT-PCR. RESULTS: Compared with CON group,blood glucose, glycosylated hemoglobin,left ventricular weight index,the content of collagen,and the expression of TGF-β_1,procollagen type I and collagen type III in left ventricular myocardial tissues of DM group were significantly increased,while the levels of blood insulin,LVEDV and SV were significantly decreased. However,compared with DM group,blood glucose,glycosylated hemoglobin,left ventricule weight index,the content of collagen,and the expression levels of TGF-β_1,procollagen type I and collagen type III in the left ventricular myocardial tissues of EGB-treated rats were significantly decreased,while the levels of blood insulin,LVEDV and SV were significantly increased. Compared with LG group,the mRNA expression levels of TGF-β_1,procollagen type I and collagen type III were significantly increased. However,compared with HG group,the mRNA expression levels of TGF-β_1,procollagen type I and collagen type III were significantly decreased after treated with EGB. CONCLUSION: EGB retards the process of myocardial fibrosis and improves the cardiac functions in type I diabetic cardiomyopathy rats by down-regulating the expression of TGF-β_1,reducing the synthesis and deposition of collagen type I and collagen type III. |
