摘要	Objective: To establish an HPLC method for the determination and study of its pharmacokinetics in Chinese volunteers. Method: The analytical column was packed with ZORBAX Extend-C18 (4.6 mm x 250mm, 5 μm), the mobile phase was acetonitrile-0.15mol/L NaH2PO4-water (45:20:35), the flow rate was 0.9ml/min, and detection wavelength was set at 256nm, respectively. Plasma samples were extracted with ethyl acetate and n-hexane. Results: The liner range of voriconazole in human plasma was 25-1600ng/ml (r = 0.9999). The lowest limit was 25ng/ml; Intra-day and inter-day RSD for the analysis of voriconazole in plasma samples at three concentration levels were all lower than 10%. Relative recoveries were 85%-115% and absolute recoveries were more than 75%, respectively. After a single dose, the pharmacokinetic parameters for voriconazole were as follows: Tmax was (1.70 ± 0.41) h, cmax was (1037.01 ± 81.18) ng/ml, t1/2 was (5.88 ± 0.93) h and AUC(0.t) was (6643.92 ± 696.70) ng·h/ml. Conclusion: The HPLC method is simple, high selective and sensitive for the determination of voriconazole in plasma.
