摘要	目的比较乌头药材不同部位毒性及总生物碱的含量差异,探讨川乌、附子二者临床应用不同的科学依据,为指导其临床应用提供参考。方法改良寇氏法测定LD50,总生物碱的含量测定按2010版药典附子项下总生物碱含量测定方法测定。结果乌头子根小鼠灌服给药的LD50值为1.7787 g/kg、须根为1.1455 g/kg、母根为1.6753 g/kg,表明须根毒性较母根、子根大,母根、子根毒性接近,总生物碱的测定结果表明须根总生物碱含量最大,母根、子根含量相近。结论生品附子和川乌毒性相近,总生物碱含量相近,提示其药效、临床功用不同可能与炮制方法不同有关;也提示药典中规定去掉须根有一定的科学依据。
其他摘要	Objective To compare the differences of toxicity and total alkaloid contents in different parts of monkshood medicinal material, and to investigate different scientific basis for clinical application of Radix Aconiti and Radix Aconiti Lateralis Preparata. Methods We used modified Karber'ss method to evaluate LD50, and determined the total alkaloid contents by pharmacopoeia method. Results LD50 of monkshood daughter roots, fibrous roots and mother tubers were 1.7787 g/kg,1.1455 g/kg,1.6753 g/kg respectively, indicating that the toxicity of fibrous roots was stronger than the other parts, and daughter roots showed similar toxicity to mother tubers. The content of total alkaloid in fibrous roots was higher than that in daughter roots and mother tubers. Daughter roots and mother tubers had the similar content of total alkaloid. Conclusion The toxicity and total alkaloid contents in Radix Aconiti Lateralis Preparata are similar to those in Radix Aconiti, which proves that different preparing methods may result in different drug effects and clinical function,in addition,it indicates that there is scientific foundation for removing fibrous roots during preparing aconite root which is required in pharmacopoeia
