摘要目的观察姜黄素对哮喘大鼠气道重塑及尾加压素Ⅱ(UⅡ)表达的影响。方法大鼠间隔7d共2次ip给以卵清蛋白(OVA)/Al(OH)3,第15天雾化吸入OVA激发,隔天1次,制备哮喘模型。按照分组,分别于雾化吸入OVA前30min,ig给予姜黄素50,100和200mg·kg-1,连续6周。图像分析技术测量支气管壁厚度和平滑肌厚度,ELISA法测定大鼠支气管肺泡灌洗液(BALF)和血清中UⅡ的含量,RTPCR检测肺组织UⅡmRNA表达。结果与正常对照组比较,哮喘模型组支气管壁厚度及平滑肌厚度均明显升高,分别增加了1.8和1.9倍(P<0.01)。与哮喘模型组比较,姜黄素组支气管壁厚度及平滑肌厚度均明显降低,分别减少了11.8%,27.8%,32.5%和16.6%,29.2%和38.4%(P<0.01)。哮喘模型组BALF和血清中UⅡ含量显著高于正常对照组,分别增加了16.9和11.9倍(P<0.01);与哮喘模型组相比,姜黄素组BALF和血清中UⅡ含量显著降低,分别减少了21.11%,50.5%,65.4%和26.92%,50.1%和60.8%(P<0.01)。与正常对照组相比,哮喘模型组肺组织UⅡmRNA表达增加了3.3倍(P<0.01);姜黄素组肺组织UⅡmRNA含量较哮喘模型组分别降低了30.3%,37.1%和52.8%(P<0.01)。结论姜黄素可以减少哮喘大鼠BALF和血清中UⅡ含量及肺组织UⅡmRNA的表达,其抑制哮喘气道重塑可能与降低UⅡ的表达有关。
其他摘要OBJECTIVE To observe the effect of curcumin on the expression of urotensinⅡ(UⅡ) in airway remodeling in asthmatic rats. METHODS The rat asthma models were sensitized with ip of 1 mg OVA with 100 μg aluminum hydroxide dissolved in 1.5 ml saline on d 1 and d 8. They were then challenged with 1% OVA aerosol every other day for 30 min from d 15 for 6 weeks. Curcumin 50, 100 and 200 mg·kg-1 was ig given 30 min before challenge from day 15 for 6 weeks, respectively. The total bronchial wall thickness and the airway smooth muscle thickness were measured by image analysis system. The contents of UⅡ in bronchoalveolar lavage fluid (BALF) and serum were detected using ELISA. UⅡ mRNA contents were detected by RT-PCR. RESULTS Compared with normal control group, the total bronchial wall thickness and the airway smooth musle thickness of asthma model group were significantly thicker (increased 2.8 and 2.9 times)(P<0.01), the total bronchial wall thickness and the airway smooth musle thickness of curcumin group were significantly thinner than in asthma model group(decreased by 11.8%, 27.8%, 32.5%, and 16.6%, 29.2%, 38.4%)(P<0.01). Compared with normal control group, the contents of UⅡ in BALF and serum of asthma model group were higher (increased 17.9 and 12.9 times, respectively)(P<0.01), while those of curcumin group were significantly lower compared with asthma model group(decreased by 21.11%, 50.5%, 65.4%, and 26.92%, 50.1%, 60.8%, respectively)(P<0.01). The UⅡmRNA contents in the curcumin groups were significantly lower than those in the asthma model group(decreased by 30.3%, 37.1%, 52.8%)(P<0.01), but those in asthma model group were higher than in normal control group(increased 4.3 times)(P<0.01). CONCLUSION Curcumin can decrease the contents of UⅡ in BALF and serum, and the expression of UⅡ mRNA of asthmatic rats, which is associated with alleviated formation of airway remodeling.
