摘要	ATP-dependent Lon protease is a nuclear DNA-encoded mitochondrial enzyme highly conserved throughout evolution,contributed to the degradation of abnormal proteins within mitochondrial matrix,in the correct folding of proteins imported in mitochondria,as well as in the maintenance of mitochondrial DNA.The expression of Lon is induced by various stimuli,including hypoxia and reactive oxygen species,which provides protection against cell stress.Lon up-regulation contributes to metabolic reprogramming in cancer cells,favors the switch from a respiratory to a glycolytic metabolism,helping cancer cell survival in the tumor microenvironment,and contributes to epithelial to mesenchymal transition(EMT).Knock-down of Lon expression,or pharmacological inhibition of its protease activity,leads to cell death in various cancer cells.Moreover,Lon could be an independent prognostic factor for overall survival of bladder cancer and colon cancer patients.Taken together,Lon protease may be included in the growing class of proteins that are not responsible for oncogenic transformation,but that are important for survival and proliferation of cancer cells,and that could serve as a potential diagnostic biomarker and therapeutic target for treatment of various cancer,as well as for prediction of the effectiveness of chemotherapy.
