摘要 | The aim of this study is to develop an ultra-high performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method to determine oxycodone in rat plasma. Diazepam was used as internal standard (IS). Rat plasma samples were processed by acetonitrile precipitation. Chromatographic separation was conducted on a UPLC BEH C18 column. The mobile phase consisted of acetonitrile and 0.1% formic acid, and the gradient elution mode was used with the total running time of 4.0 min. Electrospray ionization (ESI) source was selected. Under the positive ion mode, the quantification was performed by multiple reactions monitoring (MRM) with transition of m/z 316.1 -> 298.1 for oxycodone and m/z 285.1 -> 193.3 for IS. The results showed that in the range of 0.2-100 ng/mL, oxycodone was linear in rat plasma (r > 0.995) with the lower limit of quantification (LLOQ) of 0.2 ng/mL. The intra-day and interday precision represented as RSD of oxycodone was less than 11 and 12%, respectively. The accuracy was between 88.4 and 109.9%. The recovery was higher than 68.3% and the matrix effect was between 97.2 and 106.0%. The method is sensitive, rapid and selective and has been successfully applied to the pharmacokinetics of oxycodone in rats. |
